本劇的主角Gregory House是一名『非典型醫生』，他從不穿白大褂，永遠是鬍子拉碴，拄著一根拐杖，成天吃糖豆似的磕止痛片；

他堅信『每個人都說謊』，要么竭力避免與病人或家屬見面，要么極盡諷刺嘲弄之能，甚至對病人惡言相向，但他的話往往能一語中的，道出人們不願面對的真相；

他的診斷方式離經叛道，多番指示手下的『小鴨子』醫生破病人家門而入，尋找致病源頭；

為了解開棘手的醫學難題，他不惜採取極端的實驗性治療方案，把病人或自己逼向死亡邊緣，雖然少不免捅出漏子，但總有美女院長Cuddy和好友兼腫瘤科專家Wilson扛著。

作為FOX電視台王牌，走過第五個年頭的『豪斯醫生』劇力不減，精彩亦然。在上一季，House率領三新一舊四隻小鴨子，繼續破解醫學謎題的同時，他本人亦經歷了一系列情感危機：

季初剛與Wilson重歸於好，又陷入和Cuddy的感情拉鋸，兩人好容易有了實質進展，由於House不願改變，最終還是原地踏步。為擺脫長久的痛苦，固執的House開始尋求心理醫生和替代藥物的幫助，無奈事與願違，小鴨子Kutner意外自殺，使他陷入極端自責，以致出現嚴重幻覺，最終被關進了精神病院。

第六季將以兩小時特別版開篇，劇情從Mayfield精神病院展開，從醫生變成病號的豪斯，睿智依然，頑劣不改，為了重奪行醫執照，把醫院上下鬧得雞犬不寧。出院後，House將搬進Wilson家，兩人再度過起“同居”生活。

另一邊廂，Foreman暫代House成為部門主管，Fans們期待已久的老鴨子Chase和Cameron，亦將重新回歸主線。

在新一季中，House如何“飛躍瘋人院”，如何重操舊業？糾結的Huddy關係是前進還是倒退？ H/W同居會產生什麼化學效應？擁擠的小鴨子陣容將有何變化？
